4-Hexylresorcinol Inhibits Class I Histone Deacetylases in Human Umbilical Cord Endothelial Cells

چکیده

Histone deacetylases (HDACs) are key enzymes for post-translational modification and influence on various cellular activities. Thus, HDACs associated with many diseases their inhibitors have clinical significance. Here, 4-Hexylresorcinol (4HR) was studied as an inhibitor class I using the HDAC (HDACi) Trichostatin-A a positive control. The 4HR administered 1–100 μM to human umbilical endothelial cells (HUVECs) expression activity were examined. decreased level of HDAC1, 3, 4, 5 in time dose-dependent manner. also increased acetylated lysine significantly (p < 0.05). Collectively, new that reduced HUVECs.

Inhibitors of histone deacetylases in class I and class II suppress human osteoclasts in vitro.

Histone deacetylase inhibitors (HDACi) suppress cancer cell growth, inflammation, and bone resorption. The aim of this study was to determine the effect of inhibitors of different HDAC classes on human osteoclast activity in vitro. Human osteoclasts generated from blood mononuclear cells stimulated with receptor activator of nuclear factor kappa B (RANK) ligand were treated with a novel compoun...

FK228 (depsipeptide) as a natural prodrug that inhibits class I histone deacetylases.

FK228 is a histone deacetylase (HDAC) inhibitor, the molecular mechanism of inhibition of which has been unknown. Here we show that reduction of an intramolecular disulfide bond of FK228 greatly enhanced its inhibitory activity and that the disulfide bond was rapidly reduced in cells by cellular reducing activity involving glutathione. Computer modeling suggests that one of the sulfhydryl group...

Differentiation of Umbilical Cord Lining Membrane-Derived Mesenchymal Stem Cells into Endothelial-Like Cells

Background: Stem cell therapy for the treatment of vascular-related diseases through functional revascularization is one of the most important research areas in tissue engineering. The aim of this study was to investigate the in vitro differentiation of umbilical CL-MSC into endothelial lineage cells. Methods: In this study, isolated cells were characterized for expression of MSC-specific marke...

In-vitro Differentiation of Human Umbilical Cord Wharton’s Jelly Mesenchymal Stem Cells to Insulin-Producing Cells

Background & Objective: Diabetes is a major chronic metabolic disease in the world. Islet transplantation is a way to treat diabetes. Unfortunately, this method is restricted due to graft rejection and lack of donor islets. Mesenchymal Stem Cells (MSCS) have the ability to differentiate into Insulin-Producing Cells (IPCs). In this study, Human Umbilical Mesenchymal Stem Cells (HUMSCS) were in...

Hemogenic endothelial progenitor cells isolated from human umbilical cord blood.

Hemogenic endothelium has been identified in embryonic dorsal aorta and in tissues generated from mouse embryonic stem cells, but to date there is no evidence for such bipotential cells in postnatal tissues or blood. Here we identify a cell population from human umbilical cord blood that gives rise to both endothelial cells and hematopoietic progenitors in vitro. Cord blood CD34+/CD133+ cells p...
